DNA/RNA Extraction Reagents in Plant Research

DNA/RNA extraction is usually the first step required for most molecular biology investigations in plant research. Plants present some unique challenges compared to animal tissues in DNA/RNA extraction. For example, the tough cell wall of many plant tissues often requires time-consuming enzymatic or mechanical techniques to break apart. In addition, other problems may be encountered including the presence of large quantities of polysaccharides; high levels of RNases; various different kinds of phenolics, including tannins; low concentrations of nucleic acids (high water content); tissue, such as lignin (wood), that is difficult to break up; and so on. To overcome these problems and difficulties, TransGen provides both silica-based spin column method and modified CTAB method tailored for plant DNA/RNA extraction. Furthermore, soil DNA extraction reagent based on magnetic bead method is also available.

DNA Gel Electrophoresis Reagents in Plant Research

Agarose gel electrophoresis is the simplest method for the general analysis of plant DNA. This technique reveals the size distribution of DNA in any sample together with the approximate concentrations of the DNA components in the sample. The concentration of agarose in the gel can be varied to separate DNA molecules of different sizes. Polyacrylamide gel electrophoresis provides greater resolution than agarose gel electrophoresis in the analysis of small DNA fragments but is generally more difficult to perform. Gene Cloning Reagents in Plant Research

Cloning of PCR products is a common requirement in the production of gene constructs, useful probes and the sequencing of some PCR products. Blunt cloning has been widely used to clone PCR products, which is more suitable for blunt ended products. For products with a 3'-A overhang, vectors with a complementary T can be used to clone these products with high efficiency, which is known as TA cloning. Western Blot & Immunofluorescence (IF) Reagents in Plant Research

Western blot (WB) is a widely used immunological technique in plant research to detect specific proteins in tissue homogenate or extract. Native proteins are separated through gel electrophoresis and then transferred to a membrane, where target proteins are detected by specific antibodies. WB can detect nano-gram amount of target proteins using high quality antibodies.

Immunofluorescence light microscopy has made a significant impact on plant research over the past decade. It is a powerful yet convenient tool, as it offers all the advantages of the conventional light microscope but with increased specificity and sensitivity. It has found application in numerous investigations of plant cells, particularly those related to the plant cytoskeleton. Immunofluorescence light microscopy permits the rapid examination of the three-dimensional organization of microtubules (MTs) in fixed whole cells of plants.
