CD45, also known as leukocyte common antigen (LCA) or protein tyrosine phosphatase receptor type C (PTPRC), which consists of
single-chain type I transmembrane glycoproteins with a similar structure and a molecular weight between 180-240 kDa. CD45 is
expressed in the surface of all white blood cells (including lymphocytes, granulocytes and monocytes) except red blood cells and blood
platelets. CD45 is a tyrosine phosphorous acidase, which plays a key role in the activation mediated by antigen receptor of T and B cells
through dephosphorylation of substrates including p56Lck, p59Fyn and other Src-family kinases. In addition, CD45 is a key molecule
for signal transduction on the cell membrane, which plays a important role in the development and maturation, regulation of function
and signal transmission of lymphocytes. After purification of CD45 monoclonal antibody by affinity chromatography, the antibodies
were conjugated with fluorescein FITC under optimal conditions and used for flow cytometric detection of CD45 on cell surface.
