CD4 is a single-chain type I transmembrane glycoprotein with molecular weight of 55 kD, which is mainly expressed in helper T cells, NKT cells, monocytes, macrophages and dendritic cells etc. CD4 is a co-receptor for T-cell receptor (TCR) to recognize antigens, which binds to the non-polypeptide region of MHC class II molecules, and participates in signal transduction during TCR recognize antigen. CD4 is also the main receptor for HIV. In macrophages or NKT cells, CD4 regulates cell differentiation, activation, cytokine expression and cell migration through TCR/LCK-independent signaling pathway. After purification of CD4 monoclonal antibody by affinity chromatography, the antibodies were conjugated with fluorescein APC under optimal conditions and used for flow cytometric detection of CD4 on cell surface.
