Viral Nucleic Acid Detection Using Real Time PCR

Real-time (quantitative) PCR assays, which is now widely used to detect viral nucleic acids in clinical specimens, facilitate the very rapid, sensitive, and specific identification of many known pathogenic viruses. The genetic material of the virus can be either DNA or RNA. For DNA viruses including the adenoviruses, herpesviruses, polyomaviruses and bocaviruses, the extracted DNA can be directly detected by real time PCR. On the other hand, for RNA viruses including coronaviruses, Zika virus, dengue viruses and Ebola virus, the extracted RNA needs to be converted to DNA before detected by real time PCR or directly detected by real time RT-PCR kits.

This is a probe real time PCR kit used for virus DNA detection or virus RNA detection after reverse transcription showing the following advantages:

• Hot start with three antibodies blocking technique improves sensitivity, specificity and amplification efficiency.

• Specially optimized qPCR reaction buffer provides higher sensitivity and specificity.

• Passive reference dyes (to normalize tube-to-tube differences due to pipetting errors) are provided for different qPCR instruments, leading to more accurate results.

Application example: ASFV detection

PerfectStart II Probe qPCR SuperMix (Cat. No. AQ711) and the qPCR kit from Company TA are used for the amplification of DNA extracted from porcine serum infected with ASFV using EasyPure® Viral DNA/RNA Kit (Cat. No. ER201). The results for AQ711 and the qPCR kit from Company TA are comparable.

Application example: PRRS virus detection

PerfectStart II Probe qPCR SuperMix (Cat. No. AQ711) and the qPCR kit from Company TA are used for the amplification of DNA reverse transcribed by TransScript® One-Step gDNA Removal and cDNA Synthesis SuperMix (Cat. No. AT311) from virus RNA, which is extracted from porcine serum infected with PRRS virus using MagicPure® Viral DNA/RNA Kit (Cat. No. EC301). The results for AQ711 and the qPCR kit from Company TA are comparable.

This is a one-step probe real time RT-PCR kit used for RNA detection showing the following advantages:

·  Simple procedures reducing chance of contamination;

·  High specificity and high sensitivity leading to more accurate results.

Application example: BVDV detection

TransScript® II Probe One-Step qRT-PCR SuperMix (Cat. No. AQ321) and the qRT-PCR kit from Company TA are used for the detection of RNA extracted from cell culture medium infected with BVDV using EasyPure® Viral DNA/RNA Kit (Cat. No. ER201). The results for AQ321 and the qRT-PCR kit from Company TA are comparable.

The results for AQ321 and the qRT-PCR kit from Company TA are comparable.

This is a multiplex one-step probe real time RT-PCR kit used for RNA detection showing the following advantages:

·  Simple procedures reducing chance of contamination;

·  dUTP/ UDG used to avoid cross contamination leading to more accurate results.

·  High specificity and high sensitivity leading to more accurate results.

Application example: 2019-nCoV detection

TransScript® II Multiplex Probe One-Step qRT-PCR SuperMix UDG(Cat. No. AQ322)is used for the detection of 2019-nCoV genes (ORF1ab, N) and human internal control gene (RNase P) from 2019-nCoV-specific in vitro-transcribed RNA standard diluted by HeLa total RNA. The result shows AQ322 enables multiplex amplification with a LOD of 500 Copies/ml.
