Exosomes are nano-sized vesicles (approximately 30-150 nm in diameter) actively secreted by cells. They carry a variety of biomolecules from their parent cells, including proteins, lipids, and nucleic acids (mRNA, miRNA,lncRNA, DNA, etc.) and play key roles in intercellular communication, immune regulation, tumor microenvironment remodeling, tissue regeneration and repair. Despite the promising prospects of exosome research, researchers and industry professionals often encounter three major bottlenecks in practice:

Inconsistent Cell Culture Quality: Different culture conditions (presence or absence of serum, media formulation, GMP grade) directly affect exosome yield, composition and function, leading to irreproducible experimental results. 

Inefficient and Harsh Isolation/Purification: Traditional ultracentrifugation is time-consuming, equipment-dependent and the shear force can easily damage exosome structures; some commercial kits suffer from insufficient purity or batch-to-batch variability.

Difficulty in Downstream Nucleic Acid Analysis: Small RNA abundance within exosomes is extremely low and prone to degradation. Conventional extraction methods often fail to obtain RNA of sufficient purity, impacting downstream applications like qPCR and sequencing.

High-Quality Cell Culture System – "Source Control" for Exosomes

Exosomes can be isolated directly from various body fluids for biomarker discovery or diagnostic studies. However, when the goal is to prepare exosomes from specific cell types (e.g., functional mechanism studies or therapeutic applications of mesenchymal stem cell-derived exosomes), the quality of the donor cell culture becomes the critical starting point determining the yield, purity and functional consistency of exosomes. Mesenchymal Stem Cells (MSCs) are currently the most commonly used therapeutic cell type in exosome research. The anti-inflammatory,

pro-angiogenic and immunomodulatory active components carried by MSC-derived exosomes are closely related to the cell state. When researchers aim to engineer MSC exosomes (e.g., loading drugs or targeting ligands) or perform GMP-grade production, a standardized, exogenous interference-free cell expansion system is essential. Efficient Exosome Isolation Solutions

The isolation and purification of exosomes is a critical step determining the reliability of downstream experimental results. An ideal isolation method should satisfy: high recovery yield, high purity (low contamination from non- exosomal proteins/lipoproteins), preservation of exosome structural integrity and bioactivity, ease of operation and standardizability. TransExo® Cell Media Exosome Kit (FE401)

A product specifically for exosome extraction and purification from cell culture supernatant. The obtained exosomes exhibit high purity and high activity, suitable for various detection methods such as Western Blot, transmission electron microscopy, qPCR and particle size analysis.

Precise Small RNA Analysis Tools

After obtaining high-purity, intact exosomes, molecular analysis of their contents is central to revealing functional mechanisms, discovering biomarkers or assessing therapeutic potential. Analysis of exosome cargo focuses primarily on two levels: small RNAs (especially miRNA, tRF, piRNA) and proteins. Due to the extremely low RNA content within exosomes (typically only 0.01-0.1 fg RNA per exosome) and the significant interference from residual ribonucleoproteins and genomic DNA in samples, specially optimized extraction and analysis methods are required.

•By adjusting the volume of absolute ethanol added to the aqueous phase, the following can be obtained:

1. RNA spin column adsorbs large molecular weight RNA (28S rRNA, 18S rRNA, mRNA), and then the flow-through (containing RNAs smaller than 200 nt, such as miRNA, siRNA, shRNA, snRNA, etc.) is applied to a miRNA spin column to adsorb Small RNA.

2. RNA spin column adsorbs all RNA (including RNA smaller than 200 nt).

•Optimized ratio of poly(A) polymerase and reverse transcriptase ensures miRNA reverse transcription efficiency.
